The Future of Alzheimer’s Drug Development: Insights from SfN 2025

Video: The Future of Alzheimer’s Drug Development: Insights from SfN 2025

Alzheimer’s research is entering a hopeful new phase, moving beyond a narrow focus on amyloid towards a broader, systems-level understanding of the disease. Drawing on key insights from SfN 2025, this video explores the growing importance of synaptic dysfunction, glial and immune biology, and emerging therapeutic targets.

Video Transcript

00:03: This year’s Society for Neuroscience meeting, one message stood out: Alzheimer’s research is entering a genuinely hopeful new phase. After years of focusing narrowly on amyloid, the field is shifting toward a much broader systems-level understanding of the disease. And that shift is opening real therapeutic possibility. Three key takeaways emerged: First, synaptic dysfunction can now be viewed as one of the earliest and most actionable parts of the disease.

00:31: Tools like SV2A PET are bringing synaptic loss into focus as a measurable biomarker, which could transform how we stage patients and evaluate new therapies. Second, glial and immune biology are moving to centre stage. Microglia and astrocytes don’t just respond to pathology, they actively shape it. And new work shows that peripheral immune decline may actually drive disease progression.

00:57: This reframes Alzheimer’s as a disorder of both brain and body. Third, this broader biology is unlocking a wave of new drug targets, from synaptic eat-new signals and tau propagation machinery to immune rejuvenation approaches like PD-1 and PDR-1 modulation. Across the meeting, the town was more optimistic than it has been in decades.

01:20: The central idea is clear: Alzheimer’s isn’t a single pathway problem, and the most promising therapies ahead will integrate synaptic health, immune-brain interactions and circuit-level insight. For drug developers, this marks one of the most strategically important shifts the world has seen in years.
